Polymyxin B is a cyclic peptide antibiotic produced by Bacillus polymyxa, belonging to the polymyxin family. Its chemical structure contains lipophilic and hydrophilic groups, which enable it to effectively destroy the outer membrane of Gram negative bacteria. This drug is usually used in the form of Polymyxin B Sulfate, which appears as a white or off white powder and is easily soluble in water. Polymyxin B, due to its unique antibacterial mechanism, is widely used to treat severe infections caused by drug-resistant bacteria, especially as one of the last line of defense drugs in clinical practice.
Application&Function

Polymyxin B mainly exerts bactericidal effects by disrupting the integrity of bacterial cell membranes. Its positively charged molecules bind to lipopolysaccharides (LPS) on the outer membrane of Gram negative bacteria, leading to increased membrane permeability, leakage of intracellular substances, and ultimately bacterial death. It has strong activity against drug-resistant strains such as Pseudomonas aeruginosa, Acinetobacter baumannii, and Klebsiella pneumoniae, and is commonly used to treat sepsis, pneumonia, urinary tract infections, and burn infections. Due to the risks of nephrotoxicity and neurotoxicity, strict monitoring of blood drug concentration is required during clinical use. In addition, polymyxin B is also used in topical formulations such as eye drops and wound ointments to reduce systemic toxicity.
