Atovaquone is highly lipophilic, enabling efficient absorption when taken with fatty meals (bioavailability increases by 2–3 times). Peak plasma concentrations are achieved within 1–8 hours post-dose. The drug is extensively protein-bound (>99%) and undergoes enterohepatic recirculation, resulting in a prolonged terminal half-life of 2–3 days. It is not significantly metabolized by hepatic enzymes but is excreted unchanged in feces (~94%), minimizing drug-drug interactions. Notably, atovaquone lacks renal excretion, making it safe for patients with kidney impairment.

Application&Function

Atovaquone exerts its antiparasitic and antifungal effects by selectively targeting mitochondrial electron transport chains. In Plasmodium species, it inhibits the cytochrome bc1complex (Complex III), disrupting mitochondrial membrane potential and ATP synthesis. This action blocks the pyri1idine biosynthesis essential for parasite survival. In Pneumocystis jirovecii, atovaquone inhibits dihydroorotate dehydrogenase (DHODH), a key enzyme in pyrimidine production, leading to nucleic acid depletion and organism death.
