Ceritinib is mainly used to treat advanced or metastatic non-small cell lung cancer (NSCLC) with positive ALK gene rearrangement. Its core mechanism of action is to specifically inhibit ALK kinase activity, block downstream signaling pathways such as PI3K/AKT and MAPK, thereby inhibiting the growth, proliferation, and metastasis of tumor cells. Clinical studies have shown that Ceritinib still has significant therapeutic effects on patients resistant to crizotinib and can effectively control brain metastases. Common side effects include gastrointestinal reactions and liver dysfunction, but overall tolerability is controllable. This drug provides an important second-line treatment option for ALK positive lung cancer patients, significantly prolonging progression free survival (PFS).
