Artesunate is a potential STAT3 inhibitor with significant inhibitory effects on small cell lung cancer cell line H69, with an IC50 value of less than 5 μ M. In addition, it is also used in antimalarial research and significantly inhibits cell proliferation, induces cell apoptosis, and promotes cell cycle arrest in the G0/G1 phase in SKM-1 cells. Artesunate also has high immunosuppressive activity, low toxicity, and can inhibit T lymphocyte proliferation induced by mitogens and alloantigens
Application&Function
Artesunate is an antimalarial drug and potential anti-tumor compound, with the following core characteristics and effects:
1. Drug category and use
Antimalarial drugs: As a precursor compound of dihydroartemisinin (DHA) succinate, it exerts antimalarial effects by converting into active metabolites.
Antitumor research: demonstrated inhibitory effects on tumor cells in multiple in vitro and in vivo studies, particularly targeting small cell lung cancer, myelodysplastic syndrome (MDS), and others.
2. Pharmacological mechanism of action
Anti malaria mechanism: Inhibits the protein export protein 1 (EXP1) of malaria parasites, interfering with their metabolic processes.
Antitumor mechanism:
Inhibiting the STAT3 signaling pathway, inducing tumor cell cycle arrest (G0/G1 phase) and apoptosis;
Increase intracellular calcium ion concentration and reactive oxygen species (ROS) levels, regulate the BCL-2/BAX ratio, and reduce the expression of anti apoptotic protein survivor;
Inhibiting tumor progression through DNA demethylation, such as restoring E-cadherin expression of CDH1 gene.
In summary, Artesunate has anti malaria as its core indication and has demonstrated multi-target effects in the field of tumor therapy. Currently, related mechanism research and clinical translation are still being promoted.
