Valnemulin (pleuromutilin) is a new generation of semi-synthetic antibiotics belonging to the diterpene class, the same class as tylomycin, and is an animal-specific antibiotic. It is mainly used for the prevention and treatment of mycoplasma diseases and Gram-positive bacterial infections in pigs, cattle, sheep and poultry. It was approved by the European Community in 1999 for the prevention and treatment of swine dysentery caused by Brachyspira hyodysenteriae infection and swine endemic pneumonia caused by Mycoplasma pneumoniae infection.

Mechanism of action

The mechanism of action of Vanimulin is to inhibit the synthesis of bacterial proteins at the ribosome level and also inhibit the synthesis of RNA at high concentrations. It mainly has antibacterial properties, but it can also kill bacteria at high concentrations. It has a broad antibacterial spectrum and is effective against G+ and G- bacteria, highly effective against Mycoplasma and Spirochetes, but has relatively low efficacy against intestinal bacteria such as Escherichia coli and Salmonella.

Vorimulin was mixed feeding at 200ppm, and the drug concentration of colonic contents was 5.20ug/ml. 75ppm was mixed feeding, and the drug concentration of colonic contents was 1.68ug/ml. The concentration of colonic contents has a linear relationship with the concentration of mixed feeding.
Vanimulin undergoes extensive metabolism and excretion in pigs. Its metabolites are mainly rapidly excreted through bile and feces. 87% of the radioactive markers are excreted within 3 days after the last administration, and approximately 3% are excreted in urine during the same period.
