Pharmacological action
It can block the H1 receptor of smooth muscle, capillary wall and other tissues, so as to play a competitive antagonistic effect with histamine, and also has a significant central calming effect, and can strengthen the effects of anesthetics, hypnotics and analgesia. It also lowers body temperature and stops vomiting. Treatment of symptoms.
Pharmacology and toxicology
Promethazine is a phenothiazine derivative, an antihistamine that can be used for nausea, dizziness, motion sickness, and sedation and hypnosis.
1.Antihistamine effect: Competing with histamine released by tissues for H1 receptors, it can antagonize the contraction or contracture of the smooth muscle of the gastrointestinal tract, trachea, bronchus or bronchioles, and relieve the spasmodic and congested effect of histamine on the smooth muscle of the bronchus.
2. Antiemetic effect: It may be related to inhibiting the emetic chemosensory area of the medulla oblongata.
3. Anti-motion sickness effect: It may act on the vestibular and vomiting centers and medulloid receptors in the midbrain through central anti-cholinergic activity, mainly blocking the excitation of cholinergic synaptic labyrinth impulses in the vestibular nucleus.
4. Sedative hypnotic effect: The mechanism of inhibition of the central nervous system has not been elucidated, which may be due to the indirect reduction of the irritability of the brain stem reticular activation system.
pharmacokinetics
The absorption is fast and complete after oral or injection administration, and the protein binding rate is high. The onset time of this product is 20 minutes after oral or rectal administration, the antihistamine effect generally lasts 6 to 12 hours, and the sedation effect lasts 2 to 8 hours. It is mainly metabolized in the liver, and inactive metabolites can be excreted in the urine, with little excretion in the stool.
