Doxycycline is a semi synthetic tetracycline antibiotic obtained by deoxygenation of oxytetracycline at the 6 α - position, formulated as a hydrochloride salt. Its mechanism is the same as tetracycline, mainly interfering with the protein synthesis of sensitive bacteria. It can specifically bind to the 30S subunit of bacterial ribosomes at position A, thereby inhibiting the binding of aminoacyl tRNA at that position and preventing the elongation of peptide chains; Doxycycline can also alter the permeability of bacterial cell membranes, allowing nucleotides and other important components inside the cell to leak out, thereby inhibiting bacterial DNA replication. Doxycycline has strong antibacterial activity against Staphylococcus aureus, Streptococcus pneumoniae, Streptococcus pyogenes, Neisseria gonorrhoeae, Meningococcus, Escherichia coli, Pseudomonas aeruginosa, Shigella, Yersinia pestis, Listeria monocytogenes, etc. It also has certain effects on Rickettsia, Mycoplasma, Chlamydia, Actinomyces, etc. The antibacterial spectrum is basically the same as tetracycline and oxytetracycline. Both in vivo and in vitro antibacterial properties are stronger than tetracycline. Microorganisms have close cross resistance to doxycycline, tetracycline, oxytetracycline, and other antibiotics.
